Cole worked in an American shoe factory, where he joined the Congress of Industrial Organizations (CIO) and became an organizer for the shoe workers' union. He joined the Photo League in late 1943. There he studied with Sid Grossman, wrote for and edited Photo Notes, served as chair of the darkroom committee (1948–49), and taught the basic technique class (1950–51). As a professional photographer, he covered stories for UNICEF and International Planned Parenthood, among other human-rights and healthcare organizations. Cole's later career in photography centered on teaching. At Brooklyn College he worked first as a substitute teacher for Berenice Abbott, and then as an influential tenured professor (the school's Bernard Cole Memorial Scholarship in Photography was established in his memory). In 1974, with Milton Meltzer, he published Eye of Conscience: Photographers and Social Change.
